It was unknown if Sam and his family ever moved to Earth Colony II prior to their posting on Deneva (sans two sons), as seen in Operation -- Annihilate!. Since George "Sam" Kirk was a research biologist, it stands to reason that Earth Colony II had some form of indigenous life that may have attracted his interest.

These ships, slow by 24th century standards, were charting a quadrant that was largely unexplored. Ships, such as the Enterprise, became renowned for their discoveries. By the mid-24th century, the Federation had expanded into areas charted by these ships. For exploration into the Beta Quadrant, ships had to leave the comfort of Federation space and travel into the unknown.(6; The Sound of Her Voice; Flashback) The state of exploration in the late 23rd century was depicted in the Federation star chart "The Explored Galaxy". Many locations discovered or known to the Enterprise and her crew were located between the Tholian Assembly and Romulus. Both locations were identified in The Search, Part II as being located in this quadrant. The chart, first seen chronologically in 6, was also seen in several Star Trek: The Next Generation and Star Trek: Deep Space Nine episodes set in the 24th century, from the year 2364 to 2370. Earth Colony II was one of the Earth colonies in the 23rd century; another one was Earth Colony 5. EC II was the site for a research base where James T. Kirk's brother "Sam" hoped to move with his wife Aurelan and their three sons when Kirk last saw him – at the departure of the USS Enterprise on her five-year mission in 2265.

In the second year of the mission, on the planet Exo III, Captain Kirk tested Dr. Korby's android for precise knowledge of his personal history in the presence of Andrea, Nurse Chapel, and Korby: it knew that brother Sam hadn't said he would go to Colony II, but only that he hoped to go there. (TOS: "What Are Little Girls Made Of?")

Background

According to the Star Trek: Star Charts ("United Federation of Planets I"), in 2378, Earth Colony 2 was a Federation member.
